Possible drug interactions

Some drugs/medications may interact with Tadalafil. You should not take Tadalafil if you are taking nitrates (such as nitroglycerin tablets or patches) because the combination of these with Tadalafil could significantly lower blood pressure and lead to fainting or even death. Additionally, do not take Tadalafil if you are taking Cisapride, other ED medications (e.g. Viagra), Adcirca, or Riociguat.

Other important safety information

Patients who have a condition called “left ventricular outlet obstruction” from valvular problems or heart muscle enlargement may get side effects such as fainting or light-headedness. Because patients with recent heart attacks or stroke, heart pains (angina), heart failure, uncontrolled blood pressure or uncontrolled irregular heart beats, severe liver disease, and retinal eye problems have not been studied, Tadalafil (Cialis) is not recommended for these patients. There may be rare occurrences of priapism or painful, prolonged erections. This is a serious condition that requires immediate medical attention. Patients should seek prompt medical attention if their erection lasts longer than four hours.

Drug-Food Interactions:Do not take St. John’s wort (herbal supplement used to treat depression) while taking Dapoxetine. Avoid consuming grapefruit juice within 24 hours of taking Dapoxetine as it could lead to an increase in the level of Dapoxetine in the body.

Drug-Disease Interactions:Inform the doctor if you have cardiovascular/heart disease, a history of dizziness due to low blood pressure, depression, mania, bipolar disorder, schizophrenia, epilepsy/fits, bleeding or blood clotting problems, glaucoma, kidney problems, or moderate to severe liver problems.

Dapoxetine

Pregnancy and children's growth: Dapoxetine should not be used during pregnancy or if applicable by prescription or distribution. It should be used in women who are severely pregnant or are planning to become pregnant. Dapoxetine should not be used to treat premature ejaculation in adult women. It may increase sensitivity of semen to light. Dapoxetine should not be used during pregnancy.

Lactose intolerance: Dapoxetine can cause low blood lactase (LBL) levels in the body. Do not take Dapoxetine at the same time as lactate (Lactate level). Avoid use during pregnancy.

Liver problems: Dapoxetine is not indicated for the treatment of depression, hepatic encephalopathy (hepatitis that causes death), hepatic necrosis associated with cisapride (Nisus) and quinidine (Quinidex) in adults with schizophrenia or bipolar mania.

Furosemide (Tirrentangle): Use with caution in patients with a history of fibrinolysis (hepatitis that occurs after removing blood cells) or with alcoholic liver disease. Avoid use with ethacrynic acid (Acid in Eucalm) as this combination may cause toxicity.

Sodium (Na) the active ingredient. Avoid use while taking Dapoxetine as it may cause drowsiness. Take with food.

Potassium (K) the active ingredient. Avoid use while taking Dapoxetine as it may cause hyperkalemia (high levels of potassium in the body). Avoid use while taking potassium the active ingredient

Tricyclic antidepressants (TCAs):

Tricyclics are useful for treating psychomotor slowing (nervousness) and depression.

TCAs can enhance the effects of MAOIs (mono- and poly-2-hydroxy-4-phenyladenosine monocaratase) thereby lowering the risk of suicidal thoughts and behavior.

Dapoxetine is indicated for the treatment of major depressive disorder and obsessive-compulsive disorder. Its use in these conditions may produce unwanted effects.

In clinical trials, use of Dapoxetine, especially in the elderly, has been associated with a modest increased risk of falls and other physical injuries. Dapoxetine should not be used in children and adolescents.

It is not known if Dapoxetine is excreted in human milk. However, it is unlikely to harm children and adolescents when administered in high doses.

How Tadalafil Works

Before we explain how Tadalafil helps manage ED, we need to explain how erections occur naturally. When you become aroused, your body releases a substance called nitric oxide (NO). In turn, levels of a chemical known as cyclic guanosine monophosphate (cGMP) increase in your body, relaxing the muscles on the sides of your penis so blood can flow into the organ and trigger a stiff erection.

While all this is happening, your body also compresses the veins that take blood away from the penis, so that the blood remains in place and you can maintain that erection. Eventually, the PDE5 enzyme in your body gets to work, and it processes the cGMP. When that happens, your erection dissipates, and the penis becomes flaccid once again.

If you have ED, regardless of the underlying cause, this natural process is somehow interrupted. That makes it more difficult for you to get or maintain an erection that’s sufficient for intercourse. And that’s where Tadalafil, a PDE5 inhibitor, can help. By blocking this enzyme in your body, you’ll have an easier time getting an erection and holding on to it until sex is complete.

As we mentioned earlier, ED is caused by several factors. One of the most common culprits is diabetes. When you’re living with diabetes, your body’s ability to use blood to keep you hydrated is going to go down. That means that your body needs more blood to get to it. And that brings on an extremely high amount of cyclic guanosine monophosphate (cGMP).
